The increase in triglyceride occurred several hours after the elevation of plasma FFA levels and coincided with the development of insulin resistance(10), suggesting that an insulin resistance-causing signal was generated during the synthesis of TG. We hypothesize that diacylglycerol may be such a signal because an increase in its synthesis would coincide with that of triglycerides and because it is a well-known allosteric activator of protein kinase C(12-14), an enzyme that has been linked to insulin resistance To evaluate this possibility, DAG mass and PKC activity and distribution were assayed in skeletal muscle biopsies Because PKC is known to activate NF-kB(20,21) and because NF-kB has recently been linked to fatty acid-induced impairment of insulin action , we have also examined the possibility that the development of insulin resistance was related to alterations in the IKK/IkB/NFkB pathway.
