stem cell, 又稱為幹細胞, 其定義為只要具有differentiation以及 self-renew的能力的細胞即可稱之為stem cell.differentiation比較簡單就是細胞由一種型態轉變成另一種phenotype, 其中可能牽涉到一些新的蛋白及基因表現。stem cell的細胞分裂有兩種, symmetry cell division以及 asymmetry cell division, symmetry cell division可以增加stem cell的數量而asymmetry cell division就是一邊產生differentiated cell而另一邊產生一個活性跟原來的幹細胞活性相當的幹細胞, 這就是"self-renew"。而在幹細胞執行differentiation及self-renew時，周邊的microenvironment非常重要就是所謂的"niche"，其可以提供stem cell執行功能時所需要的養分，如neural stem cell其周邊的endothelial cell即負責這個角色! 就stem cell的function來說, 又可分為三種, pluripotent stem cell， totipotent stem cell，multipotent stem cell，在受精卵開始發育後大概 4天時,會成為一個8細胞體，此時這8個細胞每一個細胞都是totipotent stem cell ，其功能最為強大，每個細胞都能發展成一個完整的生命體!若發育到行成 blastocyst時，內層細胞團包含著pluripotent stem cell，此時每個細胞都可以發育成生命體的任何一種器官!最後一種呢，multipotent stem cell-- 就是成人幹細胞，這就不需要我多說了吧...mesenchymal stem cell， neural stem cell，HSC等等都是論文常提到的。還有一個比較特別的也是最近漸漸被注意的，就是Cancerous stem cell 我之前看的一篇PANS說到，Cancer cell為什麼經過放射腺及藥物治療後過幾天又開始活起來了??!!那篇paper提到一個論點就是cancer cell存在著 cancerous stem cell，因為此種stem cell跟一班的stem cell一樣，會表現 ABC transporter，熟的人都知道這是一種drug-resistant transporter，可以將藥物往細胞外送，減少對細胞的傷害，而那篇PNAS的論文也將Cancer裡的 cancer stem cell進而純化出來，發現他的確會造成mestasis，而普通cancer cell並不會。現在非常多的paper尤其是nature系列的常提到這種幹細胞! 喔喔今天先打這樣吧過幾天來談談 stem cell therapy 那以下是我seminar要講的paper: Hematopoietic Stem Cells Convertinto Liver Cells within Days without Fusion NATURE CELL BIOLOGY VOLUME 6 | NUMBER 6 | JUNE 2004 請多多指教摟 -- ※ 發信站: 批踢踢實業坊(ptt.cc) ◆ From: 140.114.100.165
