[求救] 請問protease種類
如題~
生產的重組蛋白一直會被切掉 His-tag
對我後續純化很不利
預測出protease的可能切位後
得到結果如下
115: LysN
116: LysC, Pepsin (pH1.3), Pepsin (pH>2), Thermolysin, Trypsin
117: Chymotrypsin-low specificity (C-term to [FYWML], not before P),
Pepsin (pH1.3), Pepsin (pH>2), Proteinase K, Thermolysin
118: Proteinase K
我只知道protease分成4大類
1. Metal protease
2 Serine protease
3. Cysteine protease
4. Aspartyl protease
想請問大家
有沒有什麼文獻或是書籍可以查詢protease的種類?
我上網找了很久得到的資訊有限
只知道LysN是屬於metal protease
Glutamyl endopeptidase, Staphylococcal peptidase I 是屬於 Aspartyl protease
如果知道這些protease屬於哪一類
我想加入相對應的protease inhibitor去試試看有無改善His-tag斷掉情形
