[求救] 做cell transfection時vector會插入chromosome嗎?
我目前在做cell transfection,並想挑出可以表現目標蛋白的stable clone.
vector用的是pcDNA4/TO/myc-His,而cell則是用293T,
transfect後有一直用抗生素zeocine來做篩選.
我想請問,
在做stable transfection時,外來的vector是否會插進host cell chromosome?
還是說,只會形成episome之類的結構,而不會插入chromosome呢?
我有先用wiki查了一下, http://en.wikipedia.org/wiki/Transfection405
上面說stable transfection時由於有用selection marker作篩選,
會讓外來基因integrate into host genome,
但我不懂這邊的"integrate into genome"是指會插入至chromosome?
還是說只是維持episome狀態,沒被踢掉而已?
