[求救] IF染membrane protein的問題
大家好
我在細胞上overexpress一個膜蛋白
抽過membrane fraction壓WB，確定他在膜上的確有高度表現
但是在染IF時遇到一個問題
我用的抗體認protein C端，但是protein是N端在膜外頭
以下是IF protocol
1. PBS wash
2. 4% PFA fix 15min at RT
3. PBS wash
4. 1% BSA blocking 1h at RT
5. primary antibody 3h at RT
6. PBS wash
7. secondary antibody 2h at RT
8. PBS wash
9. DAPI RT 10min
10. PBS wash
11. mounting
本來都沒有treat saponin，結果都看不到訊號
所以試了在step3,4之間treat 0.1% saponin，打1, 5, 10min三種condition
結果pattern是cytosol跟膜分不出來，無法看到標準的membrane pattern
找過其他家抗體，都沒有人在賣認N端的抗體
請問各位有什麼好建議?
