[求救] transfection的步驟
現在用的是santa cruz的transfection reagent和transfection medium
要用的是siRNA的系統
看到santa cruz的protocol有寫到
1.要用 antibiotic free的medium~
所以是真的不能在growth medium中加任何antibiotic嗎?
因為我家的細胞中都有加antibiotic可以直接將細胞從有antibiotic的換到沒有的嗎?
2.在transfection前，他說要用transfection medium wash過，這個步驟是有必要的嗎?
因為在transfection 過後是直接加入transtection medium+reagent+siRNA incubate
5-7hr
所以~是要照protocol寫的照做? (因為有看到說transfection前最好不要換medium)
3.這樣的做法好像是transient transfection，他是可以打下來再換到我要做實驗條件嗎?
因為第一次做，所以有以上問題想請教大家~謝謝
