qPCR NEWS - May 2012 - focus on MIQE guidelines ------------------------------------------------------------------- Dear researcher, dear Gene Quantification page reader, Our newsletter informs about the latest news in quantitative real-time PCR = (qPCR and RT-qPCR), which are compiled and summarised on the=20 Gene Quantification homepage. The focus of this newsletter issue is: * Update on the MIQE pages - http://MIQE.gene-quantification.info * New updated APP versions available in May 2012 for iOS, Android, and html= 5 - http://MIQE-press.gene-quantification.info * Follow ou new established "GeneQuan Daily NEWS" - a daily updated newslet= ter - http://Daily-News.gene-quantification.info=20 * GenEx - a powerful tool For qPCR data analysis - download a free trial ve= rsion - http://GenEx.gene-quantification.info ---------------------------------------------------------------------------= -- The MIQE Guidelines - Minimum Information for Publication of Quantitative R= eal-Time PCR Experiments=20 Stephen A. Bustin, Vladimir Benes, Jeremy A. Garson, Jan Hellemans, Jim= Huggett, Mikael Kubista, Reinhold Mueller, Tania Nolan, Michael W. Pfaf= fl, Gregory L. Shipley, Jo Vandesompele, and Carl T. Wittwer Clinical Chemistry 2009, 55(4): 611-622 http://www.gene-quantification.de/miqe-bustin-et-al-clin-chem-2009.pdf BACKGROUND: Currently, a lack of consensus exists on how best to perform a= nd interpret quantitative real-time PCR (qPCR) experiments. The problem is = exacerbated by a lack of sufficient experimental detail in many publication= s, which impedes a reader's ability to evaluate critically the quality of t= he results presented or to repeat the experiments. CONTENT: The Minimum Information for Publication of Quantitative Real-Time= PCR Experiments (MIQE) guidelines target the reliability of results to hel= p ensure the integrity of the scientific literature, promote consistency be= tween laboratories, and increase experimental transparency. MIQE is a set o= f guidelines that describe the minimum information necessary for evaluating= qPCR experiments. Included is a checklist to accompany the initial submiss= ion of a manuscript to the publisher. By providing all relevant experimenta= l conditions and assay characteristics, reviewers can assess the validity o= f the protocols used. Full disclosure of all reagents, sequences, and analy= sis methods is necessary to enable other investigators to reproduce results= .. MIQE details should be published either in abbreviated form or as an onli= ne supplement. SUMMARY: Following these guidelines will encourage better experimental pra= ctice, allowing more reliable and unequivocal interpretation of qPCR result= s. http://MIQE.gene-quantification.info The MIQE guidelines has been frequently cited by other researchers: =3D> 7= 54 times until May 2012 =3D> http://scholar.google.com/scholar?cites=3D6338124712618390161&as_sdt= =3D2005&sciodt=3D0,5&hl=3Den ---------------------------------------------------------------------------= -- updated APP versions for iOS, Android and html5 MIQE qPCR APP for iOS - iPhone, iPad and iPod 25th May 2012 - A neew updated version is available! For the first time ever, checkl= ists are optimized in real time: you can reach 100% for every project if yo= u are MIQE compliant. Checklists are specific for each project type: just click on the kind of nu= cleic acid you are working with, the checklists adapts instantly by removin= g unnecessary items (Reverse transcription items are not relevant if workin= g only on DNA for instance). Moreover, some items may not apply to your specific experiments. You can no= w remove them and have the most accurate MIQE compliancy. Built in= a modular fashion GenEx provides a multitude of functionalities for the qP= CR community, ranging from basic data editing and management to advanced cu= tting-edge data analysis. View our webpage =3D> http://GenEx.gene-quantific= ation.info Basic data editing and management Arguably the most important part of qPCR experiments is to pre-process the = raw data into shape for subsequent statistical analyses. The pre-processing= steps need to be performed consistently in correct order and with confiden= ce. GenEx Standard=92s streamlined and user-friendly interface ensures mist= ake-free data handling. Intuitive and powerful presentation tools allow pro= fessional illustrations of even the most complex experimental designs. Advanced cutting-edge data analysis When you need more advanced analyses GenEx Enterprise is the product for yo= u. Powerful enough to demonstrate feasibility it often proves sufficient fo= r most users demands. Current features include parametric and non-parametri= c statistical tests, Principal Component Analysis, and Artificial Neural Ne= tworks. New features are continuously added to GenEx with close attention t= o customers=92 needs. New features Sample handling and samples individual biology often contribute to confound= ing experimental variability. By using the new nested ANOVA feature in GenE= x version 5 user will be able to evaluate variance contributions from each = step in the experimental procedure. With a good knowledge of the variance c= ontributions, an appropriate distribution of experimental replicates can be= selected to minimize confounding variance and maximize the power of the ex= perimental design! For experiments with complex features, such as for examp= le multifactorial diseases, analytical relationships and classifications ma= y not readily be available. The support vector machine feature in the new v= ersion of GenEx is so easy to use that it will make this advanced supervise= d classification method easily available to novice users, while providing a= ccess to advanced parameters for experts.
