[求救] Invertrd invasion assays
我曉得Matrigel invasion assay的方法
但是Inverted invasion assay我一直看不懂
以下是paper上敘述的方法:
Matrigel (BD biosciences) or collagen (BD biosciences), supplemented with
25 μg/ml fibronectin (FN, Sigma) was polymerized in transwell inserts
(Corning) for 45 minutes at 37°. The inserts were inverted and 3-5x105
cells were seeded at the base of the insert. The inserts were placed in
serum free medium and the upper chamber was filled with 10% FCS and
25ng/ml EGF (Sigma) or 1U/ml TGFβ (Preprotech).
我不懂的是將insert反過來後
細胞要怎麼種在insert上?
然後insert又要放進serum free medium內
細胞不會再掉出來嗎?
