首先先對positive selection和negative selection下定義
為什麼要有positive 和negative selection
positive selection主要刪除"無法"辨認self MHC
如果無法辨識self MHC 則alpha beta T細胞則會進行
heavy chain和 light chain editing(這兩個算是補救措施)或是apoptosis
negative selection則是針對會對self MHC+self peptide會有強烈反應的T cell做刪選
一旦會有強烈的反應就會產生activation induced death
就是透過Fas FasL類似的路徑來讓T細胞apoptosis
所以negative selection是刪除對自體會產生傷害的T cell
當然Central tolerance(thymus bone marrow)也不是這麼神 也會有些漏網之魚
所以還要靠Peripheral tolerance(secondary lymph organ)來幫忙
